SPECIFICATIONS.
Chromatographically purified, salt-free, lyophilized, water soluble powder.
Single band on SDS gel electrophoresis at 28 KD.
Optimum pH 8.8.
Free of trypsin and chymotrypsin as well as other pancreatic enzymes.
Store 5°C. Stable 9-12 months.
3.2 - 3.8 units per mg of protein. One unit will hydrolyze 1µmole of Suc-(Al)3-pNA per minute at pH 8.8 at 37°C.
ASSAY.
Prepare 1 mg/ml in 50 mM NaOAc pH 5 containing 200 mM NaCl.
E, 1%, 280nm = 10.51.
A280 x 0.95 = mg/ml.
Use 5 to 10 µgm in the assay buffer; 100 mM Tris pH 8.8 containing 0.2 mM Suc-(Al)3-pNA at 37°C.
ACTIVITY.
