Since ES12 is a mixture of peptides Mr 3000-60000 it runs as a smear on PAGE. Usually no distinct bands are seen.
Elastin-Soluble is prepared from Elastin No. E60 by the method of Partridge, S.M., Davis, H.F., and Adair, S.T., Biochem, J., 61, 11 (1955). In the method, bovine neck ligament elastin is solublized with hot oxalic acid solution, clarified, diaylzed and lyophilized. The preparation is a salt-free, water soluble powder that readily forms a coacervate at pH 5.0 and 37°C.
Elastin-Soluble may be used to determine elastolytic activity by the method of Keller. S., and Mandl, I., Biochem. Med., 5, 342, (1971). In the method, elastolytic material is incubated at pH 8.8 with Elastin-Soluble. Released peptides are quantitatively determined with ninhydrin reagent. Small quantities of pure elastase (0.5 to 5 mgm) are specifically detected without interaction by non-elastolytic protein. The technique may be used to determine the elastolytic inhibitory activity of serum.

b) The elastin soluble was derived only from animals which were slaughtered in the United States in USDA inspected abattoirs where the animals were subjected to ante- and post mortem inspection and the animals were fit for human consumption or use in laboratory studies.
