SPECIFICATIONS.

Affinity purified from human plasma.  Lyophilized from 0.05 M Tris pH 7.5 + 0.1M NaCl.  Reconstitute with H2O.  Store at 5°C.

 

PROTEIN CONCENTRATION.

(A280 x 1.45) - (A260 x 0.74) = mg/ml               

 

SPECIFIC ACTIVITY.

One unit will hydrolyze 1mmole of furylacrylol-alanyl-lysine (F-Ala-Lys) per minute at pH 7.8 at 37°C. 

The substrate-buffer is 0.05 M MOPS pH 7.8, 0.5 M NaCl, 0.35 mM F-Ala-Lys. 

Π=  -1.2 of 1 mM F-Ala-Lys at 336 nM.

Note 1.  The substrate-buffer should be pre-incubated at 37°C for at least 5 minutes to reduce background absorbance (336nm).

 

Note 2.  For 1 mM F-Ala-Lys, the -∆Abs for totally hydrolyzed is

approximately  -1.0 to -1.2 at 336 nM in 10 mm cell.  Preparations may vary in Î and should be determined at 336 nM.

 

The human plasma has been tested and found to be nonreactive for hepatitis B surface antigen (HVsAg), human immunodeficiency virus type 1 antigen (HIV Ag), antibodies to hepatitis B core (HBc), hepatitis C virus (HCV), human immunodeficiency virus type 1 and type 2 (HIV-1/2) and the human T-cell lymphotropic virus type I (HTLV-I); to have alanine aminotransferase (ALT) levels acceptable for transfusion; and to have negative test results for the serologic test for syphilis (STS).
